Ecotropic Receptor Booster

Ecotropic receptor booster technologies are used to temporarily elevate the density of viral receptor proteins on the surface of any target cell that you wish to infect. The technology allows you to efficiently transduce cells that are resistant to viral infection due to low receptor density. The Ecotropic Receptor Booster, for example, allows you to efficiently transduce human cells with ecotropic retrovirus or lentivirus that otherwise can only infect rodent cells.

Receptor booster technologies are used to temporarily elevate the density of viral receptor proteins on the surface of any target cell that you wish to infect. The technology allows you to efficiently transduce cells that are resistant to viral infection due to low receptor density. The Ecotropic Receptor Booster allows you to efficiently transduce human cells with ecotropic retrovirus or lentivirus that otherwise can only infect rodent cells.

How does it work?

Each receptor booster consists of a vial of concentrated exosome-like vesicles that are coated with the mCAT-1 receptor protein. When the booster vesicles are applied to your cells (10–20 µl per well of a 6-well plate for ~2 hours), they fuse with the plasma membrane, resulting in an increased level of the receptor on the cell surface. Your target cells can then be more efficiently transduced with ecotropic retrovirus.

The Ecotropic Receptor Booster increases the transduction efficiency of cells that are either resistant to infection, or inefficiently infected by ecotropic pseudotyped lentivirus or retrovirus. The Ecotropic Receptor Booster consists of exosome-like particles coated with the mCAT-1 receptor. When applied to target cells, fusion of the exosome-like particles with the host cell plasma membrane temporarily increases surface mCAT-1 receptor, thus allowing for efficient viral transduction.The Ecotropic Receptor Booster renders cells that cannot usually be transduced with ecotropic psuedotyped virus (e.g. human celllines) susceptible to viral infection.

Receptor boosters are concentrated exosome-like vesicles that are applied to your target cells prior to infection with virus. Boosters result in increased transduction efficiency because treatment increases the cell surface density of the receptor recognized by the lentivirus (ecotropic) or retrovirus (ecotropic or amphotropic). Using this technology, for example, you can coat human cells with the ecotropic receptor (which is otherwise absent), enabling them to be transduced with ecotropic retrovirus or ecotropic lentivirus.

This kit provides a complete retroviral vector packaging system for use with any retroviral vector. It is designed to produce virus that can infect dividing cells from a broad range of mammalian and nonmammalian cell types. The GP2-293 Packaging Cell Line contains only the MoMuLV gag and pol genes. The viral envelope portion of the packaging function (env gene) is supplied by transiently co-transfecting one of the Retro-X Universal Packaging Vectors (p10A1, pAmpho, pEco, or pVSV-G) with the retroviral vector. The pQCLIN Retroviral Vector (included in the vector set) can be used as a positive control to constitutively express β-galactosidase in transduced mammalian cells.

High titer retroviral packaging with a choice of four different retroviral packaging envelopes. GP2-293 Cells were transfected with a Retro-X Q vector expressing the red fluorescent protein DsRed2, and each of the four envelope vectors supplied with the Retro-X Unversal Packaging System. After incubating the transfected GP2-293 cells for 48 hr, 10 ?l of the resulting virus was used to infect NIH 3T3 cells. After 48 hours, titers were measured by flow cytometry. Titers were calculated by multiplying the percent infected cells (infectious units) by the total cell number, then dividing by the volume of virus (ml). Panel A. Ecotropic envelope. Panel B.
