High-throughput total RNA isolation from blood—NucleoSpin 8/96 RNA Blood

High-throughput isolation of total RNA from blood

The NucleoSpin 8/96 RNA Blood kits combine the stringency of guanidine-isothiocyanate lysis with effective digestion of genomic DNA by rDNase treatment and the speed of silica membrane technology, allowing purification of total RNA from up to 400 µl of whole blood. These kits offer consistent RNA quality, reproducible yields of RNA, and high-throughput procedures free of cross-contamination.

The NucleoSpin 8/96 RNA Blood kits combine the stringency of guanidine-isothiocyanate lysis with effective digestion of genomic DNA by rDNase treatment and the speed of silica membrane technology, allowing purification of total RNA from up to 400 µl of whole blood. These kits offer consistent RNA quality, reproducible yields of RNA, and high-throughput procedures free of cross-contamination.

In the NucleoSpin 8/96 RNA Blood procedure, blood cells are directly lysed by incubation in a solution containing large amounts of chaotropic ions. This lysis buffer immediately inactivates RNases and creates appropriate binding conditions which favor adsorption of RNA to the silica membrane of the provided binding strips/plates. Contaminating DNA, which is also bound to the silica membrane, is efficiently removed by on-column DNA digestion (a 15-min digestion at room temperature is sufficient, due to the highly specific activity of the rDNase). Salts, metabolites, and macromolecular cellular components are removed by simple washing steps with three different buffers. Pure RNA is eluted under low ionic strength conditions with RNase-free water.

The NucleoSpin 8/96 RNA Blood kits provide a powerful tool for diverse research applications, including gene expression profiling, drug discovery, and development of novel molecular diagnostics.

Effective DNA removal

Effective DNA removal. Total RNA was isolated manually under vacuum from 400-µl whole blood samples (EDTA) according to the standard protocol for NucleoSpin 96 RNA Blood, with and without an on-column rDNase treatment. A PCR for human elongation factor EF-1a was performed to determine the amount of residual genomic DNA in the eluted RNA using a LightCycler system and gene-specific primers designed to yield a 469-bp product. The figure shows the corresponding amplification curves of the 0.01 ng–100 ng DNA standards (dark blue lines), the NTC (negative control; green), and replicates of the blood sample 1 (1-1, 1-2), with DNase treatment (+ DNase; red lines) and without DNase treatment (-DNase; light blue lines). In total, blood samples from six individual donors were analyzed by PCR (data not shown). Samples treated with rDNase (+DNase) show an average Cp of 31.8. Samples purified without rDNase (-DNase) treatment show an average Cp of 25.9. Omitting the on-column DNA digestion results in an approximately 64 times higher amount of residual gDNA.
